If the hitchlock cannot, under any circumstances, interfere with the overrun brake, yes, but, to be on the safe side, I would leave it off.
 
I believe they are designed that they can be used when hitched, however I've also heard/read stories that emergency services don't like them as they can prevent quick separation of the van and tow vehicle in the event of an accident. I never do and would only consider using it when stationary, at services etc, not that I ever have.
